We are looking for a Research Technician or Senior Research Technician to provide technical support for the Structural Biology lab. The lab is interested in developing a new generation of optical imaging technologies to enable advances in two hitherto difficult-to-investigate areas: the real-time analysis of macromolecular interactions and motions at the nanometer scale in vivo and the three-dimensional architecture of complex molecular machines.
Responsibilities include, but are not limited to:
*Preparation of mammalian nuclear factors, including recombinant proteins produced in bacteria and insect cells, as well as native large macromolecular complexes from human cells
*Assay purified proteins using in vitro transcription assays, native gel electrophoresis, DNAse I and permanganate footprinting
*Design, clone, and assay constructs for various genes of interest using standard molecular and cell biology techniques (PCR, transformation, cloning, subcloning, DNA/RNA miniprep and maxiprep, Western and Northern blotting, etc.)
*Responsible for cell culture procedures, including large-scale growth of mammalian cells in suspension and of mouse embryonic stem cells
*Ensure stocks of lab supplies are maintained, place orders as necessary, maintain lab equipment and perform other duties as assigned
*BA/BS in biological sciences with a minimum of 1-3 years research experience. Master's degree highly preferred.
*Strong technical ability in molecular/cell biology and biochemistry required
*Good verbal and written communication, interpersonal, and computer skills required
*Familiarity with gene targeting techniques, preferably in mice
*Previous protein purification experience preferably with endogenous nuclear complexes
*Ability to be organized, dependable, efficient, and to work independently once fully trained
